Will this actually work on a 5.56 rifle because .223 and 5.56 are different sizes

.223 and 5.56 are NOT different sizes. The difference in the two loadins has to do with the pressure they are loaded to and the additional throating in the the 5.56 chamber. The cartridges have the same physical size.

Load the bullet bore sight in the breach, you can close or keep open then point the barrel with the red dot at however many yards you plan to shoot out and adjust level and windage to about dead center of your scope. I usually will put my cross hairs about 1moa up at 25-50yards which that should almost be dead nuts at a hundred yards.

Does the laser need to be adjusted or is it accurately leveled from the factory?

Mine is accurate from the factory.....I inserted the laser and set the sights, then rotated the laser in the bore 90 deg. and checked the gun site again...I did this for the other 2 90 deg. rotations. Looked to be accurate enough to head for the range. I hope this helps.

Can you practice trigger pull without damaging the laser sight?

No this is just a bore sighting tool. For adjusting sights. Scopes and such. I also have a laser training cartridge for the purpose of dry fire trigger practice. Do a search for laser training cartridge. That's what you want for that purpose.
