Can I use them for my fluval 306?

It will fit any 16ml to 22ml plastic pipe you can use any filter with that type of pipe

I'm guessing that you want to leave the hoses in the tank and effectively separate the filter for maintenance. If that is why then you will need two separate pairs. If you haven't already bought the filter, have a look at the Eheim Pro 3 filters. The have a dual isolation built in so you just close the valve and press a button to separate from the hoses. The valve stops the tank siphoning out and just clicks back in after you perform maintenance / cleaning of the unit. They are bigger and cost more but they really are excellent.

Will this connect directly to the actual plastic outlet pipe and then join to the flexible hose?

No. Just use a small piece of 12/16 mm ehiem hose then connect the two

What is the difference between double tap and single tap?

Single taps only seal one side of the connector, double taps seal both sides of the connection. So, if you close both ends no water can escape from either side of the hose. For instance, if you had a chiller for your tank and put a double on both the inlet and outlet, you could disconnect the chiller without removing the hoses from the tank and none of the water in the chiller could escape when you move it. With a single, one side let's water escape from the side without a valve when you disconnect.
