PI want to hang a 5 foot round macrame swing to hold at least 600 pounds. I want to install on 2 inch side of wood (of a 2 by 6) on a pergola.

The only issue I see is that the mounting screws are about three or possibly four inches long,so they will go through your two inch thick piece of wood and into whatever it is attached to. Also, I would use something closer to four inches thick to mount it to just due to the amount of stress. If you put 600 lbs, or anything near that heavy on a swing it will generate a lot more than 600lbs of force as it swings or spins. I dont have the formulas on me to be exact but 600 lbs of dead weight would generate closer to 1000 lbs of force swinging from a height of about 6 to 10 feet high. The hanger is rated for that amount so as long as you mount it properly you should be fine.

The length of the lag bolts is 3 1/3 Inch.

will 2" rope fit through the shackle?

No, The Shackle is not nearly big enough to accommodate that size rope. Assuming you are using this for a climbing rope you will have to use something like a large D-link chain link to connect it to the shackle.
