Would you be allowed this and a case with wheels as hand luggage on an airplane

FYI, the external dimensions of the 40L backpack are 55x34x18 cm (21.6x13.4x7.1 inch).

Can this be taken on as a cabin bag or hand luggage?

FYI, the external dimensions of this bag is 55x34x16cm (21x13x6 Inch). It depends on the regulations of different airline.

Is this allowed as a 'small bag' on Ryanair (i.e. without having to pay as carry on luggage)?

I'm using it on ryan air all the time and I never had any issues. Be sure you don't have a second bag, eg purse or duty free bag.

Will this fit an a3 folder in it?

Upon checking, the standard dimension of A3 folder is 42 x 29.7 cm. It could be put in the 17 inch backpack easily.
