Can you do this in a live tree?

YOu need to cut a freshly living tree but the tree should not still be living as its natural immune defence will try to kill the oyster mycelium.

Would these work in a coffee grounds system instead of a log?

They are mushroom spores stuck to a wooden dowel. I can't speak to a coffee substrate specifically but I don't see why these wouldn't work in any acceptable substrate including a sawdust pack. Logs just offer a larger food source and will result in a larger and longer supply of mushrooms for several years while a less dense substrate would work but offer a lower harvest.
