What is the material of the spatula? What kind of plastic is it made of?

Just a normal silicon plastic like a spatula in your kitchen.

What's the difference between CPU rest on a lines or dots of thermal paste with pressure vs spreading?

The main thing is to make sure the thermal paste spreads evenly and isn't too thick or too thin. I personally don't do the "DOT" or the 'line". I do a thin layer over the entire surface (how I was taught in the Air Force) so I know how thick it is before adding my cooler.
