Is this a Shimano brand or is it a counterfeit knockoff ?

Sure “looks” like the real deal. Mine arrived without original/shimano packaging but the levers calipers and hoses all branded shimano. These arrived in 3 weeks much to my surprise.

Can these work on a Specialized Enduro 2001?

You might need an adaptor to mount the brakes on a frame that old. My brakes came with all the adaptors I needed.

Will it fit on a giant roam

As long as you are using 160 rotors you should be okay.
