I only use the Munkcare swabs to brush my cat's teeth. According to the ingredients on the Munkcare box, it states that those swabs are sugar free and are NOT made with rubber latex. I have been using Munkcare swabs to brush my cats teeth for several years, with very good results.

Is it okay to put toothpaste on them

I don’t see why you can’t put toothpaste on it. However, as long as the person can spit it out.

Will the sponge come off when use ?

if the sponge is forced thru opening they can come off
