I didn't wait very long at all and it worked for me.

Once I connect the tube the printer head, it is basically sealed, no fluid can possibly pass through, what am I missing?

The fluid should dissolve the clog as you slowly force it through the head with the syringe. It worked for me. If it's not working, your clog may be too solid. Try letting it sit for a while.
